不同基质对六堡茶茶树品系扦插繁育的影响

Cutting Propagation of Liupao Tea Plant with Various Substrates

    目的 为解决六堡茶茶树品系快速繁育问题,筛选适宜培养六堡茶茶树品系的基质,对推广六堡茶茶树品系扦插繁育技术提供一定的科学依据。
    方法 以4个六堡茶茶树品系(梧茶12、梧茶16、梧茶17、梧茶18)为试验材料,采用短穗扦插方法,研究黄泥(作为对照)、V黄泥∶V锯木屑(2∶1)、V泥炭土∶V珍珠岩(5∶1)、V泥炭土∶V珍珠岩∶锯木屑(4∶1∶2)4种处理对六堡茶茶树品系的愈伤组织形成率、生根率、成活率和出圃率的影响。
    结果 不同处理对六堡茶茶树品系的愈伤组织形成率、生根率、成活率和出圃率均有明显影响。扦插30 d,处理1(黄泥∶锯木屑=2∶1)愈伤组织形成率最高;扦插40~60 d,处理2(泥炭土∶珍珠岩=5∶1)和处理3(泥炭土∶珍珠岩∶锯木屑=4∶1∶2)愈伤组织形成率显著高于对照处理(P<0.05);扦插70~80 d,3种处理愈伤组织形成率较高于对照处理。扦插90 d,处理2(泥炭土∶珍珠岩=5∶1)生根率最高,3种处理对梧茶12生根率为100%。扦插260 d,3种处理的成活率和出圃率极显著高于对照处理(P<0.01),着叶数高于对照处理。
    结论 综合基质和茶树品系考虑,梧茶12在处理2(泥炭土∶珍珠岩=5∶1)基质上的生根率、成活率和出圃率最好,分别达100%、90.67%、83.33%。

     

    Abstract:
    Objective Substrate for rapid propagating Liupao tea strains using plant cuttings was explored.
    Method Cuttings with short spikes from four Liupao tea strains, i.e., Wucha 12, Wucha 16, Wucha 17, and Wucha 18, were cultured on loess (CK) and volumetric mixtures of loess∶sawdust at a ratio of 2∶1 (T1), peat soil∶perlite at 5∶1 (T2) or peat soil∶perlite∶sawdust at 4∶1∶2 (T3). Rates of callus formation, rooting, survival, and qualified plant count of the explants in the propagation experiment were determined.
    Results Significant impacts on the physiological indicators of the cuttings cultured on the substrates were observed. Thirty days after start of cultivation, CK produced the greatest rate on callus formation among the various substrates. However, in 40-60 d, the rate became significantly higher with T1 and T2 (P<0.05). By the time of 70-80 d of cultivation, the lowest rate was found on the cuttings grown on CK. On rooting, the cuttings showed the highest rate on T1 by the 90th day, and Wucha 12 explants cultured on all mixture substrates had a perfect rating of 100%. In 260 d, the survival rate and qualification rate of the propagated plants cultivated on the 3 mixture substrates were significantly higher than the control treatment (at P<0.01), and the number of leaves was higher than those on CK.
    Conclusion Among the tested Liupao tea strains and culture substrates, T2, which applied a mixture of peat soil and perlite in a volumetric ratio of 5∶1, as a substrate for cultivating Wucha 12 explants rendered the highest rates of rooting at 100%, survival at 90.67%, and 83.33% plants qualified for transplantation. It was recommended for the cutting propagation program.
