外源激素对‘中茶108’插穗生根、氧化酶活性及内源激素的影响

Rooting, Enzyme Activities, and Endogenous Hormones of Zhongcha 108 Cuttings Affected by Plant Regulator Application

    目的 为提高‘中茶108’基质扦插的生根效果,并研究其生根过程中酶活性和内源激素的变化规律。
    方法 研究不同种类、质量浓度及其组合的外源激素对‘中茶108’插穗生根情况的影响,分光光度法测定吲哚乙酸氧化酶(Indole acetic acid oxidase, IAAO)、过氧化物酶(Peroxidase, POD)和多酚氧化酶(Polyphenol oxidase, PPO)3种氧化酶活性,ELISA检测吲哚乙酸(Indole acetic acid, IAA)、脱落酸(Abscisic acid, ABA)和玉米素核苷(Zeatin riboside, ZR)3种内源激素含量。
    结果 外源激素萘乙酸(1-Naphthaleneacetic acid, NAA)和吲哚丁酸(Indole-3-butyric acid, IBA)处理均可促进‘中茶108’插穗生根。当NAA质量浓度为50~150 mg·L−1 时,随着其质量浓度的升高,插穗生根效果越好。150 mg·L−1 NAA处理的生根率、根鲜重、最长根长和平均根长最高,分别为89.82%、0.18 g、8.26 cm和3.78 cm;生根数以50 mg·L−1 NAA与50 mg·L−1 IBA等体积组合的处理最高,为10.80条;与CK(清水处理)相比均达到显著差异(P<0.05)。外源激素的施用使IAA含量在处理后1~15 d升高,峰值达24.60 ng·g−1;同时,IAAO活性在1~10 d出现下降,内源ABA含量在1~15 d始终低于对照组,从而生根。
    结论 在本试验条件下,150 mg·L−1 NAA浸泡2 h为‘中茶108’最佳扦插条件。

     

    Abstract:
    Objective Rooting, enzyme activities, and endogenous hormones of Zhongcha 108 stem cuttings were studied to determine the applicability of root growth promotors for plant propagating.
    Methods Different types of growth regulators in varied concentrations were applied on Zhongcha 108 stem cuttings to determine the effects on rooting and plant physiology. Activities of indole acetic acid oxidase (IAAO), peroxidase (POD), and polyphenol oxidase (PPO) of the plantlets were detected by spectrophotometry, and contents of endogenous indole acetic acid (IAA), abscisic acid (ABA), and zeatin riboside (ZR) by ELISA.
    Results Application of exogenous hormones, 1-naphthaleneacetic acid (NAA) and indole-3-butyric acid (IBA), promoted rooting of the cuttings. Although increasing NAA concentration brought about greater root growth, the application at 150 mg·L−1 rendered the highest growth rate of 89.82% and fresh root weight of 0.18 g, as well as 8.26 and averaging 3.78 longest roots per cutting. The applied regulators raised the content of IAA and ABA with a declined activity of IAAO in the tissue which stimulated root growth.
    Conclusion By applying 150 mg·L−1 NAA on Zhongcha 108 stem cuttings, rooting of the plantlets was significantly enhanced to facilitate the propagation program.
